Bead example image is available from examples folder under "hologram_example.tif"
- Get the example image by downloading the latest source code through releases. You can skip this if you're using your own example.
- Open the app. Click "Select File" under "Processing" section.
- Navigate to your chosen file location. We currently support tif or tiff only.
- After opening the file, click "Set ROI". A new window will open with chosen image. Select region of interest for retrieval by clicking and dragging. The image and ROI will appear in the preview panel on the lower right.
- Click either "Run Manual" or "Run Auto" to start the phase retrieval process. We suggest "Run Manual" for finer control. 5.a) "Run Manual" - A new panel will appear with the Fourier transform of the image. Three separable sections should be seen in the image. Click and drag to select the region on the right side of the image (this may be on the upper or lower quadrant depending on your image). 5.b) "Run Auto" - "Upper Limit" and "Noise Level" can be used to control thresholding process.
- Once complete, the program will display 6 figures showing the phase and intensity of the image. This can be saved and manipulated as desired.
- Click Start Preview to initiate camera.
- Click Live Recon.
- In the new window, click and drag to select region of interest.
- In the new window, click and drag to select the first order.
Click and drag to select the area on the right side of the FFT, as shown in image below:
For quantitative volume from phase, the settings at the bottom of "Processing" must be correctly entered based off your optical setup.
